Les projets de recherche développés par Vanessa et son équipe "Facteurs de transcription et destin cellulaire" visent à mieux comprendre la plasticité et la stabilité des phénotypes cellulaires en analysant la génération de la diversité neuronale au sein de la moelle épinière embryonnaire et la transformation qui mène aux rhabdomyosarcomes, des cancers pédiatriques. Elles se concentrent tout particulièrement sur les facteurs de transcription PAX dont l’activité modulaire est capable de générer une incroyable diversité des états cellulaires.

Titre: A BMP mediated switch in PAX3/7 transcriptional activity to pattern the sensory spinal cord
Résumé:
The emergence of cellular diversity in the developing central nervous system is controlled by the combined activity of secreted signals and transcription factors. The mechanisms by which neural progenitors integrate these two signals have yet to be deciphered. To this end, we are studying the interaction between bone morphogenetic proteins (BMPs) and the transcription factors PAX3 and PAX7 during the specification of relay (RIn) and associating (AIn) interneurons present in the dorsal embryonic spinal cord. Using mouse embryos and pluripotent stem cell-derived spinal organoids, we show that PAX proteins regulate the generation of IR and IA differently. For (AIn), PAXs bind to cis-regulatory genomic regions near key ventral specifiers where they participate in the deposition of the repressive histone mark H3K27me3, preventing progenitors from acquiring a ventral identity. This repressive activity is also present in IR progenitors. However, in these progenitors, PAX can also act as transcriptional activators, opening and activating enhancers, nearby genes essential to IR differentiation. Finally, we have demonstrated that BMP signaling in IR progenitors modulates the transcriptional potential of PAX, switching them into activators and increasing their specification potential tenfold.

Perfectionnement de la communication cellulaire – le modèle d’une synapse du cervelet

Dans ce séminaire, je présenterai la ligne directrice de mon approche en physiologie jusqu’à aujourd’hui : l’étude quantitative des communications entre cellules nerveuses par des approches en temps réel. Je me suis notamment penché sur les interactions métaboliques neurone-glie, le rôle des transporteurs membranaires et les signalisations synaptiques glutaminergiques et cannabinoïdes. Mon modèle d’étude privilégié a été une synapse excitatrice centrale, la synapse entre la cellule en grain et la cellule de Purkinje du cervelet. Cette synapse est apparue avec les vertébrés. En terme d’évolution, il s’agit peut-être du site de communication le plus perfectionné du monde vivant. Le cervelet est en effet une structure cérébrale dont le volume s’est accentué avec le développement du cerveau chez les mammifères. Chez l’homme, plus de la moitié des neurones s’y trouvent. La synapse grain-Purkinje y est prépondérante. Elle est considérée comme le siège d’une plasticité dont de nombreux mécanismes ont été décrits. Pourtant, à ce jour, ceux qui sous-tendent l’apprentissage cérébelleux ne peuvent être considérés comme établis. Un fait l’illustre : il semble qu’une large majorité de ces synapses, bien qu’en apparence normales structurellement, ne transmettent pas de dépolarisation postsynaptique : elles sont dites silencieuses. Or le rôle de ces synapses silencieuses reste à déterminer. Leur nombre varie-t-il avec l’apprentissage cérébelleux ? Ou sont-elles une constante homéostatique du réseau ? Dans quelles conditions peuvent-elles être rendues fonctionnelles ? Mes travaux récents ont abordé ces questions. Plus généralement ils s’inscrivent dans une perspective visant à une compréhension du fonctionnement synaptique qui nécessite de distinguer les mécanismes les plus fondamentaux de ceux les plus raffinés.

Biological memory: from epigenetics to polymer formation
Complex systems can be successfully modeled in computer simulations. Due to the high computational cost of the numerical algorithms, different techniques are adapted to suit particular time- and length scales. Consequently, molecular reactions and conformational dynamics of molecules are most often not represented simultaneously, which is in contrast with complex molecular systems, where the motion and chemical reactivity of molecules are coupled. Here I present a technique that is a hybrid between stochastic dynamics and kinetics and that can represent both the spatio-temporal evolution and chemical reactivity in biological and chemical systems. Modeling of two classes of systems will be presented; (I) the spreading of epigenetic modifications on chromatin so as to model the establishment of gene expression patterns that account for phenotypic diversity of multicellular organisms, (II) the formation of informational sequences such as the DNA.

Titre : Cell cycle and functional maturation in a model of human beta pancreatic cells, the EndoCbetaH2 cell line
Abstract :
In humans and rodents, pancreatic beta cells, which produce Insulin, undergo a post-natal maturation characterized by cessation of proliferation, senescence and essential functional changes. The recently described human beta cell line, termed EndoCbetaH2, was generated through lentivirus-mediated transfer of two oncogenes, hTERT and SV40LT, in fetal pancreatic cells (1). EndoCbetaH2 cells display reversible immortalization : upon removal, or silencing, of hTERT and SV40LT, EndoCbetaH2 cells exit from cell cycle, deepen their beta cell traits and show functional changes, in a process akin to post natal maturation of « true » pancreatic beta cells (1-4). To study and monitor both cell cycle and terminal maturation of pancreatic beta cells, we first generated and characterized a Fucci (Fluorescent Ubiquitination-based Cell Cycle Indicator) derivative of EndoCbetaH2 cells (2). Using this tool, we next reported the role of the RB tumor suppressor family, mostly of RB itself, in the « identity » of human pancreatic beta cells, and in their terminal maturation (3).
Chez les rongeurs et chez l’Homme, les cellules beta du pancréas, qui produisent l’insuline, subissent une maturation fonctionnelle post-natale associée à un arrêt de la prolifération et la sénescence. Récemment, une lignée de cellules pancréatiques beta humaines, la lignée EndocBetaH2, a été obtenue grâce au transfert de deux oncogènes, hTERT et SV40LT, dans des cellules de pancréas foetaux (1). Cette lignée présente une immortalisation réversible : l’excision ou l’extinction des deux transgènes immortalisants y provoque la sortie du cycle cellulaire, l’exacerbation de nombreux traits caractéristiques des cellules beta et des changements fonctionnels (1-3). Ce processus ressemble d’assez près à la maturation des « vraies » cellules beta après la naissance (4). Afin de mieux comprendre et d’étudier le cycle cellulaire et la maturation des cellules pancréatiques beta humaines, nous avons tout d’abord généré et validé une lignée dérivée portant les deux indicateurs Fucci (Fluorescent Ubiquitination-based Cell Cycle Indicator) du cycle cellulaire à partir des cellules EndoCbetaH2 (2). Avec cet outil, nous avons ensuite montré le rôle essentiel des protéines de la famille RB, P130 (RBL2) et surtout RB lui-même, dans le maintien de « l’identité » des cellules beta humaines et dans leur maturation fonctionnelle (3).
